Effective remediation of polluted environments is a priority in both Eastern and Western countries. In the U.S. and Europe, remediation costs generally exceed the net economic value of the land. As a result, scientists and engineers on both sides of the Atlantic have aggressively tried to develop novel technologies to meet regulatory standards at a fraction of the costs. In situ remediation shows considerable promise from both technical and economic perspectives. In situ technologies that deploy natural attenuating agents such as humic substances (HS) may be even more cost effective. Numerous studies have shown humics capable of altering both the chemical and the physical speciation of the ecotoxicants and in turn attenuate potential adverse environmental repercussions. Furthermore, the reserves of inexpensive humic materials are immense. Which suggests HS portend great promise as inexpensive amendments to mitigate the environmental impacts of ecotoxicants and as active agents in remediation. To elucidate emerging concepts of humics-based remediation technologies, we organized the NATO Advanced Research Workshop (ARW), entitled "Use of humates to remediate polluted environments: from theory to practice", held on September 23-29, 2002 in Zvenigorod, Russia (see the web-site http://www.mgumus.chem.msu.ru/arw).

Over the past decade, interest in plant biostimulants has been on the rise, compelled by the growing interest of researchers, extension specialists, private industries, and farmers in integrating these products in the array of environmentally friendly tools to secure improved crop performance, nutrient efficiency, product quality, and yield stability. Plant biostimulants include diverse organic and inorganic substances, natural compounds, and/or beneficial microorganisms such as humic acids, protein hydrolysates, seaweed and plant extracts, silicon, endophytic fungi like mycorrhizal fungi, and plant growth-promoting rhizobacteria belonging to the genera Azospirillum, Azotobacter, and Rhizobium. Other substances (e.g., chitosan and other biopolymers and inorganic compounds) can have biostimulant properties, but their classification within the group of biostimulants is still under consideration. Plant biostimulants are usually applied to high-value crops, mainly greenhouse crops, fruit trees and vines, open-field crops, flowers, and ornamentals to sustainably increase yield and product quality. The global biostimulant market is currently estimated at about $2.0 billion and is expected to reach $3.0 billion by 2021 at an annual growth rate of 13%. A growing interest in plant biostimulants from industries and scientists was demonstrated by the high number of published peer-reviewed articles, conferences, workshops, and symposia in the past ten years. This book compiles several original research articles, technology reports, methods, opinions, perspectives, and invited reviews and mini reviews dissecting the biostimulatory action of these natural compounds and substances and beneficial microorganisms on crops grown under optimal and suboptimal growing conditions (e.g., salinity, drought, nutrient deficiency and toxicity, heavy metal contaminations, waterlogging, and adverse soil pH conditions). Also included are contributions dealing with the effect as well as the molecular and physiological mechanisms of plant biostimulants on nutrient efficiency, product quality, and modulation of the microbial population both quantitatively and qualitatively. Pyrene, a four-ring polycyclic aromatic hydrocarbon (PAH), was identified as the chemical that requires the largest land area for soil bioremediation due to the slow rate of biodegradation at the Libby, Montana Superfund site. Prepared bed land treatment is the specific bioremediation technology that is currently employed at this site. Although bioremediation has been widely accepted for treatment of contaminated soil due to its low cost, the effective application of bioremediation is often hindered by the lack of information related to: 1) biochemical pathways, 2) enzymatic mechanisms, and 3) effects of amendments. Mycobacterium sp. KMS is a new strain isolated from the land treatment units of the Libby site and has been found to utilize pyrene as a carbon and energy source. The genome of Mycobacterium sp. KMS was sequenced by Joint Genome Institute (JGI) and is publically available in the NCBI database. This dissertation is comprised of seven chapters. Chapter I provides information concerning PAH characteristics, the Libby Superfund site, accelerated bioremediation approaches, and the hypotheses for this dissertation. Chapter 2 addresses the pyrene degradation pathway used by Mycobacterium sp. KMS based on isolating and identifying pyrene degradation intermediates. Chapter 3 describes the enzymatic mechanism of pyrene degradation by Mycobacterium sp. KMS. Chapter 4 presents the effect of Elliott soil HA (ESHA) amendment on pyrene solubility in soil slurry systems and pyrene mineralization in unsaturated soil microcosms. Chapter 5 describes the overall effect of ESHA amendment on pyrene distribution in a soil slurry system. Chapter 6 addresses the engineering significance of this study and future research recommendations. Chapter 7 summarizes the dissertation. The main theme of this dissertation is to provide the basic scientific information and knowledge for better understanding, better control, and improvement of the bioremediation process at PAH-contaminated sites.
