A NATO Advanced Study Institute on "Membrane Receptors, Dynamics and Energetics" was held in order to consider recent developments in membrane receptor research. Presentations and discussions focussed on signal transduction mechanisms (e.g. 2 G-proteins, transducin, Ca +) and on the structure and func tion of some selected receptors (i.e. low density lipoprotein receptor, acetylcholine receptor, growth factor receptors, toxin receptors). These topics were put in the larger con text of current knowledge on the structure and function of membranes; connections between different fields of research were established by in-depth discussions of energy transduc tion and transport mechanisms. Specialized membrane systems were discussed including chloroplasts and chromaffin granules. Functional relationships with specific lipids (e.g. phospho inositides, lipid A) were major topics of discussion. This book presents the content of the major lectures and a selection of most relevant posters presented during the course of the Institute. This book is intended to make the proceedings of the Institute accessible to a larger audience and to offer a comprehensive account of those topics of re ceptor, membrane and bioenergetics research that were at the center of interest during our stay on the Island of Spetsai from August 17 to 30, 1986.

Free energy constitutes the most important thermodynamic quantity to understand how chemical species recognize each other, associate or react. Examples of problems in which knowledge of the underlying free energy behaviour is required, include conformational equilibria and molecular association, partitioning between immiscible liquids, receptor-drug interaction, protein-protein and protein-DNA association, and protein stability. This volume sets out to present a coherent and comprehensive account of the concepts that underlie different approaches devised for the determination of free energies. The reader will gain the necessary insight into the theoretical and computational foundations of the subject and will be presented with relevant applications from molecular-level modelling and simulations of chemical and biological systems. Both formally accurate and approximate methods are covered using both classical and quantum mechanical descriptions. The school held at Villa Marigola, Lerici, Italy, in July 1997 was very much an educational experiment aimed not just at teaching a new generation of students the latest developments in computer simulation methods and theory, but also at bringing together researchers from the condensed matter computer simulation community, the biophysical chemistry community and the quantum dynamics community to confront the shared problem: the development of methods to treat the dynamics of quantum condensed phase systems.This volume collects the lectures delivered there. Due to the focus of the school, the contributions divide along natural lines into two broad groups: (1) the most sophisticated forms of the art of computer simulation, including biased phase space sampling schemes, methods which address the multiplicity of time scales in condensed phase problems, and static equilibrium methods for treating quantum systems; (2) the contributions on quantum dynamics, including methods for mixing quantum and classical dynamics in condensed phase simulations and methods capable of treating all degrees of freedom quantum-mechanically.
