In recent years there have been many technical developments in immunology that have potential or actual applications in many other branches of the life sciences. These developments include a number of ultrasensitive systems for the detection and assay of a wide range of biological materials, including micro-organisms and their products, as well as techniques for isolating and purifying micro-organisms and other antigenic materials. Two developments in particular have greatly extended the scope and applicability of immunological methods. These are the production of monoclonal antibodies and the introduction of enzyme linked immunoassay as a simple, safe and economical alternative to radioimmunoassay. This book is based on information gathered at the Society for Applied Bacteriology Autumn Demonstration Meeting held at the South Bank Polytechnic, London on October 15th, 1986.

Immunological Methods a compendium of basic research techniques being used in one of the largest immunology research institutes, the Basel Institute for Immunology, with particular emphasis given to new methodology. The procedures have been described by individuals judged to be highly expert in their specialties. In many instances the methods developed or adapted to unique uses by the contributors have not previously been described in detail. Techniques such as isotope laboratory maintenance; chemical modification of proteins, haptens, and solid supports, and haptenation of viable biological carriers; production of antisera against allotypes and histocompatibility antigens and production of antibody with clonai dominance; histocompatibility and MLR testing; and cell separation by haptenated gels and by velocity sedimentation of rosette-forming cells are also discussed. Other chapters cover detection of antibody-secreting and alloantigen-binding cells; immune responses in vitro and their analysis by limiting dilution; production of T-cell factors; hybridoma production by cell fusion; maintenance of cell lines and cloning in semisolid media; and the mathematical analysis of immunological data.

Moreover, it underlines the close relationship between diagnosis, therapy, and epidemiology. An outstanding example of these connections is given by the recent development of hepatitis B vaccine. The discovery and identification of a non cultivable agent by physicochemical and immunological methods were the heralds of a new era in the prevention of infectious diseases. This book provides an up-to-date, comprehensive review of developments and future aspects in various fields. The current diagnostic methods for the great variety of microbial agents affecting health are clearly unsatisfactory. New important pathogens have emerged including the agent responsible for bovine spongiform encephalopathy. Moreover, there is an increasing need for more accurate microbial control of our environment, and of the food and water we consume. What is needed are rapid, sensitive and reliable procedures which, on the one hand, should be suitable for automation and, onthe other hand, presented in a cost-effective version suitable for field use. Including new biochemical approaches, such as polymerase chain reaction, recombinant gene products and synthetic peptides, these needs are discussed in these protocols of the RAMI-90 congress.
